摘要 [目的]比较8种黄酮对血小板聚集的抑制作用并探讨其构效关系。[方法]采用体外实验方法,测定黄酮类化合物对ADP诱导大鼠血小板聚集的抑制作用,并计算其抑制率。[结果]槲皮素、芦丁、异槲皮素、橙皮素、柚皮素、橙皮苷、柚皮苷和淫羊藿苷对血小板聚集的抑制率分别为(68.33±2.43)%、(55.34±2.09)%、(52.27±4.65)%、(50.80±7.03)%、(33.07±2.09)%、(31.28±4.65)%、(22.91±1.68)%和(20.94±3.20)%,8种黄酮类化合物对血小板聚集均呈现抑制作用。与对照组(不加黄酮类化合物)相比,差异均有显著性意义(P<0.05)。[结论]黄酮类化合物对血小板聚集有明显抑制作用,A环C7位和C环C3位的羟基是该类化合物抑制血小板聚集发挥作用的重要功能基团。 [Objective] To compare the inhibitory effects of eight flavonoid phytochemicals on platelet aggregation and also explore the structure-function association.[Methods] The inhibitory effects of eight flavonoids on ADP-induced rat platelet aggregation were studied in vitro and obtained their inhibiting percentages.[Results] The results indicated that all the eight flavonoids including quercetin,rutin,isoquercitrin,hesperetin,naringenin,hesperidin,naringin and icariin could inhibit the ADP-induced rat platelet aggregation by(68.33±2.43)%,(55.34±2.09)%,(52.27±4.65)%,(50.80±7.03)%,(33.07±2.09)%,(31.28±4.65)%,(22.91±1.68)% and(20.94±3.20)%.[Conclusion] The apparent inhibitory effects of flavonoids on platelet aggregation are potentially associated with their intrinsic structures.The 7-OH group of A ring and 3-OH group of C ring were important for its antiplatelet function.
