摘要
2-氨基-4,5-二甲基噻吩-3-甲酸乙酯1与七乙酰基乳糖基异硫氰酸酯2在干燥的乙腈溶液中发生亲核加成反应,生成七乙酰基乳糖基硫脲中间体3.后者在甲醇/甲醇钠催化条件下,同时发生环化和酯交换反应,生成水溶性良好的3-乳糖基-5,6-二甲基-4-羰基-噻吩并[2,3-d]嘧啶-2-硫醇钠4目标化合物.
The nueleophilic additional reaction of ethyl 2-amino-4,5-dimethyl-thiophene-3-carboxylate 1 with hetpa-O-acetyl-lactosyl isothiocyanate 2 produced the betpa-O-acetyl-lactosyl thiourea intermediate 3 in anhy- drous solution of acetonitrile. At the CH3OH/CH3ONa condition, the later was underwent cyclization and ester exchanged reaction simultaneously, and was converted to the target compound sodium 3-Lactosyl-5,6-dim- ethyl-4-oxo-thieno[2,3-d]pyrimidine-2-thiolate 4, which had good solubility in water.
