红归胶囊治疗鼻咽癌的离体实验 被引量:2

摘要 目的:研究红归胶囊对人鼻咽癌细胞菌株CNE2的抑制和诱导凋亡的干预作用,并揭示其机制。方法:采用MTT、TUNEL、流式细胞术(FCM)、ELISA和实时荧光定量PCR(qPCR)法检测不同浓度红归胶囊对人鼻咽癌细胞CNE2的生长、凋亡、细胞周期及对血管内皮生长因子(VEGF)和环氧化酶-2(COX-2)蛋白和基因表达影响。结果:红归胶囊作用后CNE2细胞抑制率明显增加,且呈浓度及时间依赖性,以浓度100μmol·L-1时效果最好,96h抑制率为80.35%,与空白组比较有统计学意义(p<0.01);有明显诱导凋亡的作用(p<0.01);用药后细胞周期G0/G1期,S期,G2/M细胞增殖分别受到抑制;COX-2、VEGF蛋白和COX-2mRNA、VEGFmRNA的表达减少。结论:红归胶囊具有抑制鼻咽癌细胞增殖和诱导细胞凋亡的作用,其抗癌机制可能与抑制COX-2、VEGF的表达有关。 Objective: To evaluate the surpressing and inducing apoptosis effect of Honggui capsule on human nasopharyngeal carcinoma cell line CNE2, and to analyze the possible mechanism. Methods: We used MTT, TUNEL, Flow cytometry (FCM), ELISA and Real-time quantitative (qPCR) to measure the inhibition rate, apoptosis ratio, cell cycle and the expression of COX-2 and VEGF protein and mRNA of different concentration-rations of Honggui capsule on CNE2. Results: It was shown that the inhibition rate of CNE2 cell was significantly increased by Honggui capsule in concentration and time-dependent manners. The peak value (80.35%) reached at 100 ~a mol/L Honggui capsule for 96h, and the inhibition rate was significant differences compared to control group ( P 〈0.01). Honggui capsule could also induce apoptosis ( p 〈0.01). The cell cycle was arrested at G0/G1, S, G2/M. The results of EISA and RT-PCR showed that Honggui capsule reduce the expression of COX-2 and VEGF. Conclusion : Honggui capsule can inhibit the proliferation CNE2 cells and induce apoptosis, and its mechanism maybe relate with inhibiting the expression of COX-2 and VEGF.
