摘要
(S)-2-哌啶甲酸在微波辅助下,依次与氯化亚砜反应20 min、2,6-二甲基苯胺反应30 min制得(S)-N-(2,6-二甲基苯基)-2-哌啶甲酰胺(5)。5再与溴丙烷在DMF中,经微波辅助进行N-丙基化反应,制得盐酸罗哌卡因,总收率约56%。
Ropivacaine hydrochloride was synthesized from piperidin-2-carboxylic acid by successive reaction with SOC12 for 20 rain and 2,6-dimethylaniline for 30 min under microwave irradiation to give (S)-N-(2,6- dimethylphenyl) -piperidin-2-carboxamide (5), which was subjected to N-propylation under microwave irradiation with an overall yield of about 56%.
