摘要
目的观察癌痛消颗粒对移植性肝癌模型大鼠肝癌及癌旁组织微小核糖核酸(miRNAs)表达的调节作用,探讨癌痛消颗粒抗肝癌作用的机制。方法将60只移植性肝癌模型大鼠随机分为5组,即癌痛消颗粒高、中、低剂量组及5-氟尿嘧啶(5-Fu)对照组、蒸馏水对照组,另设10只大鼠为空白对照组,不进行造模及给药。于造模后第8 d对癌痛消颗粒高、中、低剂量及蒸馏水对照组灌胃给药治疗,连续用药14 d后停药;5-Fu对照组腹腔注射给药治疗,共连续注射5次,各组均停药24 h后处死各组大鼠,取出瘤块,用实时定量PCR(PT-PCR)技术观察肝癌及癌旁组织特定miRNAs中miR-199a-3p、miR-199a-5p、miR-18表达水平。结果癌痛消颗粒高、中、低剂量组及5-Fu对照组、蒸馏水对照组肝癌及癌旁组织miR-199a-3p、miR-199a-5p、miR-18表达水平与空白对照组比较差异有统计学意义(P<0.05);癌痛消颗粒高、中、低剂量组及5-Fu对照组miR-199a-3p、miR-199a-5p、miR-18表达水平与蒸馏水组比较差异有统计学意义(P<0.05),且癌痛消颗粒高、中剂量组与5-Fu对照组比较差异有统计学意义(P<0.05),癌痛消颗粒低剂量组与5-Fu对照组比较差异无统计学意义(P>0.05),癌痛消颗粒中剂量组与高剂量组比较差异有统计学意义(P<0.05)。结论癌痛消颗粒对移植性肝癌模型大鼠的肝癌及癌旁组织miRNAs的表达具有调节作用,其抗癌机制可能与降低miR-18的表达,升高miR-199a-3p、miR-199a-5p的表达有关,且癌痛消颗粒的中剂量组的临床效果最好。
Objective To observe the regulation function of Aitongxiao granule on miRNAs expression of hepatocellular carcinoma and adjacent tissue in transplanted liver cancer rat model. Methods 60 transplanted liver cancer model rats were randomly divided into five groups, high - , middle - , low - dose Aitongxiao granule group, 5 - Fu control group and distiller water group. Another 10 rats were used as a blank control group, without the model- ing and administration. At 8th day after modeling rats in high - , middle - , low - dose Aitongxiao granule group, and distiller water group received garage treatment of continuous medication for 14 days. Rats in 5 - Fu control group received intraperitoneal injection they. Rats in each group were sacrificed after 24 h of withdrawal, tumor mass was removed. The levels of miR - 199a - 3p, miR - 199a - 5p and miR - 18 of miRNAs expression in hepatocellular car- cinoma and adjacent tissue were detected by PT - PCR. Results The levels of miR - 199a - 3p, miR - 199a - 5p and miR -18 expression in high - , middle -, low -dose Aitongxiao granule group, 5 -Fu control group and distill- er water group were differ from those in blank control group ( P 〈 0.05). There was statistically significance betweenhigh-, middle-, low- dose Aitongxiao granule group, 5 - Fu control group and distiller water group on miR - 199a -3p, miR - 199a - 5p and miR - 18 expression levels ( P 〈 O. 05). There was no significant difference between low - dose Aitongxiao granule group and 5 - Fu control group on miR - 199a -3p, miR - 199a -5p and miR - 18 expression levels ( P 〉 0. 05 ). There was significant difference be- tween high-, middle- dose Aitongxiao granule group and5 - Fu control group on miR - 199a - 3p, miR - 199a - 5p and miR - 18 expression levels ( P 〈 0.05 ). Conclu- sion Aitongxiao granule has the regulation function miRNAs expression of hepatoeellular carcinoma and adjacent tis- sue in transplanted liver cancer rat model. Its anticancer mechanism may be associated with decreased expression of miR - 18 increased miR - 199a - 3p, miR - 199a - 5p expression. And middle dose Aitongxiao granule group has best clinical effect.
