缬沙坦对高脂模型大鼠动脉粥样硬化及炎性因子的影响 被引量:5

摘要 目的研究缬沙坦对高脂模型大鼠动脉粥样硬化及炎性因子水平的影响。方法通过建立高脂血症动脉粥样硬化大鼠模型,动态、系统地监测血脂及高敏C反应蛋白(hs-CRP)、肿瘤坏死因子(TNF)-α、白介素(IL)-6和IL-8的水平,同时采用Trizol法提取RNA,用RT-PCR法测量主动脉中hs-CRP mRNA、TNF-αmRNA和IL-6 mRNA的表达,并且观察粥样硬化动脉内膜形态学的改变。结果缬沙坦药物治疗后,血清总胆固醇(TC)、低密度脂蛋白胆固醇(LDL-C)水平较高脂模型组有所降低,但差异无统计学意义(P>0.05)。高脂模型组的hs-CRP、TNF-α、IL-6、IL-8均升高,且与正常对照组比较差异具有统计学意义(P<0.05)。应用缬沙坦治疗10 w后,hs-CRP和IL-6的水平均降低,与高脂模型组比较差异有统计学意义(P<0.05);TNF-α和IL-8的水平均降低,但与高脂模型组比较差异无统计学意义(P>0.05)。应用缬沙坦治疗10 w后,hs-CRP mRNA,TNF-αmRNA和IL-6 mRNA表达均降低,与高脂模型组比较差异有统计学意义(P<0.05)。缬沙坦药物干预组内膜病变较高脂模型比较有所减轻。结论缬沙坦能够有效地降压的同时,能减少部分炎性因子释放的作用,发挥其抗动脉粥样硬化的作用。 Objective To explore effects of valsartan on inflammation factors and atherosclerosis in rats model with hyperlipaemia. Methods Rat model of hyperlipaemia and atherosclerosis was established. Levels of serum lipids, CRP, TNF-α, IL-6, IL-8 were detected before and after the intervention of valsartan. The mRNA expressions of hs-CRP, TNF-α, IL-6 in aorta were measured by RT-PCR method. Aorta structures were observed under light microscope by hematoxylin-eosin staining. Results The levels of serum total cholesterol (TC) and low density lipoprotein cholesterol (LDL-C) in valsartan group were lower than those of hyperlipaemia group, but there were no significant difference ( P 〉 0. 05 ). Serum levels and mRNA expressions of hs-CRP, TNF-α, IL-6, IL-8 in hyperlipaemia group were significantly higher than those of normal group ( P 〈 0. 05 ). The expressions of hs-CRP and IL-6 mRNA in valsartan group were lower than those of hyperlipaemia group, there were significant difference between valsartan and hyperlipaemia groups ( P 〈 0.05 ). The expressions of TNF-α and IL- 8 in valsartan group were lower than those of hyperlipaemia group, but there were no significant difference ( P 〉 0. 05 ). At the end of 10 weeks, the mRNA expression of hs-CRP, TNF-α, IL-6 in valsartan group were lower than those of hyperlipaemia group, and had significant difference between valsartan and hypedipaemia groups ( P 〈 0. 05 ). Valsartan group had different levels of improvement in endometrial lesions compared with hyperlipaemia group. Conclusions Valsartan can effectively lower blood pressure and reduce the role of some inflammatory cytokine, it will play its anti-atherosclerotic effect.
