摘要
目的观察血必净对急性肺损伤(ALI)兔模型核因子-κB(NF-κB)信号通路的影响及其对ALI的治疗作用。方法选择新西兰兔80只,按随机方法分为四组:空白对照组、单纯急性肺损伤(LPS)组、单纯血必净(XBJ)组、血必净治疗组(LPS+XBJ),并给予相应的治疗,于12、24、36、48 h分批处死后分离肺组织后,采用凝胶电泳迁延率法(EMSA)加计算机辅助图像分析技术检测白细胞NF-κB活性。结果与空白对照组相比较,LPS组各时间点肺组织NF-κB表达量均明显增高(P<0.01);与LPS组相同时间点比较,LPS+XBJ组24、36、48 h肺组织NF-κB表达量明显减少(P<0.01);与空白对照组比较,XBJ组各时间点肺组织NF-κB表达量有一定减少,但差异无统计学意义(P>0.05)。结论 NF-κB参与了急性肺损伤过程,血必净能抑制NF-κB的表达,从而对肺组织起到保护作用。
Objective To observe the effects of Xuebijing on NF-κB pathways in acute lung injury(ALI) rabbits.Methods 80 New Zealand rabbits were selected and randomized into 4 groups,including the control group,LPS group,XBJ group and LPS+XBJ group,which were treated with corresponding therapies.The rabbits were killed in batches at 12th,24th,36th,48th hour and the lung tissues were reselected for further examinations.The bioactivity of NF-κB in white cells was detected by electrophoretic mobility shift assay(EMSA) in combination with computer assisted graphical analysis.Results Compared with the control group,the LPS group showed significantly higher expression of NF-κB at different time points(P 0.01);compared with the LPS group,LPS+XBJ group showed significantly lower expression of NF-κB at 24th,36th and 48th hour(P 0.01);compared with the control group,the XBJ group showed lower expression of NF-κB at different time points,however,there was no significant difference between the two groups(P 0.05).Conclusion NF-κB plays an important role in the development of ALI and Xuebijing can inhibit the expression of NF-κB,taking protective effects on lung tissues.
