摘要
目的探讨多药耐药基因GST—πmRNA在胃癌组织中的表达及临床意义。方法采用逆转录-聚合酶链反应(reverse transcription—polymerasec haint eaction.RT-PCR)技术检测53例新鲜胃癌组织标本(实验组)和15例正常胃黏膜组织(对照组)中GST-π基因的表达.并分析其表达与胃癌病理特征之间的关系。结果①53例胃癌组织中GST-πmRNA的表达(0.56±0.27)要高于15倒正常胃黏膜组织的表达(0.18±0.13)(t=5.30.P〈0.001)。②GST-πmRNA的表达与胃癌的分化程度之间差异具有统计学意义(t=2.67.P=0.01),而与患者性别、肿瘤部位、大小之间、是否淋巴结转移差异无统计学显著性意义。结论①GST-πmRNA的高表达可能是胃癌产生多药耐药的机制之一;②GST—π的检测对临床制定化疗方案有一定指导意义。
Objective To investigate the expression levels of GST-π mRNA in patients with gastric cancer and clinical signifi- cance. Methods RT-PCR method was used to detect the expression levels of GST-II mRNA in fresh gastric cancer 53 cases (experiment group) and 15 cases of normal gastric mucosa tissue(eontrol group),analyzed the relationship between the expression of GST-π and the clinicopathologic parameters. Results (1) The expression level of GST-nmRNA (0.56 ± 0. 27) was significantly higher in gastric cancer tissues than that in normal tissues (0. 18±0.13) (t=5.30,P-(0. 001). QThe expression of GST-π intensity was related to differentiation degree(t= 2.67. P= 0.01), had nothing to do with other clinico- pathologic parameters. Conclusion OThe over-expression levels of GST-πm RNA would underline the muhidrug resistance of gastric cancer;(2)The detection of expression can be looked as an importance method for judging the prognosis in gastric cancer.
