摘要
目的 研究体外培养及成纤维细胞生长因子对骨髓基质细胞生长的影响。方法 抽取兔骨髓基质细胞体外培养 ,倒置显微镜动态观察 ,从原代培养 (P0 )中观察到细胞集落的形态有明显差异 ,提示骨髓基质细胞是一个由多种细胞组成的混合体。在培养液中加入碱性成纤维细胞生长因子 (bFGF ,5ng/ml)。 结果 第一代细胞(P1)的生长速度增至对照组的 1.5倍。而未经bFGF处理的细胞活跃增殖能力只能保持 4~ 5代 ,随后即发生退化 ;而经bFGF处理的细胞 ,传 10代以上仍能保持活跃的增殖能力。结论 bFGF能促进骨髓基质细胞增殖并长期保持其生物活性。
Objective The effect of bFGF on the proliferation of BMSCs cultured in vitro is studied in this article.Method The BMSC scultured in vitro with or without the treatment of bFGF,the cell morphology and the speed of cell growth are observed under reserved microscope.Result Without the treatment of bFGF,the BMSCs degenerated after 4~5 passages;Treated with bFGF,the speed of cell growth increase 1.5 times,and the cells remains their bioactivity for more than ten passages.Concluslon Treating with bFGF facilitates the proliferation of BMSCs,and keep their bioactivity over a long period of time.
