同种异系抗原门静脉接种诱导特异性免疫抑制的研究

Study on abrogation of the capacity of delayed-type hypersensitivity responses to alloantigens and suppression of mixed lymphocyte reaction by portal venous inoculation with allogeneic cells

摘要 目的 探讨同种异系抗原经门静脉接种后机体对该抗原免疫反应状态的改变。方法 以NIH/q小鼠为供体 ,BALB/C小鼠为受体。预先用NIH/q小鼠脾细胞或与供受体无关C5 7/BL小鼠脾细胞注射至BALB/C小鼠门静脉或下腔静脉 ,1周后用NIH/q小鼠脾细胞注射至BALB/C小鼠背部皮下免疫 ,2周后第 2次皮下免疫。第 2次皮下接种后 1周 ,测定供受体之间的混合淋巴细胞反应 (MLR)和受体对供体抗原的迟发性超敏反应 (DTH)。结果 供体脾细胞门静脉接种组供受体混合淋巴细胞反应 (MLR)其刺激指数 (SI)为 3.5 1± 0 .5 5 ,DTH脚掌厚度增值为 (7.41± 0 .6 )× 10 -2mm ,而供体脾细胞下腔静脉接种组以及与供受体无关的C5 7/BL小鼠脾细胞门静脉接种组供受体MLR(SI)分别为 34.83± 3.90和 35 .33± 2 .5 4,DTH脚掌厚度增值分别为 (2 3 .5 1± 1.6 9)× 10 -2 mm和 (2 3 .87± 2 .76 )× 10 -2 mm ,差异有显著性 (P <0 .0 5 )。结论 同种异系抗原门静脉接种可诱导机体产生对该抗原的特异性免疫无反应 ,其作用具有抗原特异性。 Objective\ To study the change of the level of immunoreactions to allogeneic cells after receiving portal venous inoculation with allogeneic cells in mice. Methods\ Splenocytes from NIH/q or third-party C57/BL spleens were injected into BALB/C hosts via the host portal vein or the systemic vein. Seven days later, BALB/C mice were inoculated on the dorsal flanks with NIH/q mice splenocytes depleted of erythrocytes. Mixed lymphocyte reaction (MLR) of BALB/C to NIH/q mice alloantigens was investigated and anti-NIH/q delayed-type hypersensitivity (DTH) responses assessed by directly challenging NIH/q splenocytes in a volume of 25μl into BALB/C hind footpads 7 days later. Results\ Portal venous inoculation of NIH/q splenocytes significantly abolished the capability of the animals to develop MLR responses and DTH responses to NIH/q alloantigens as induced by subcutaneous immunization with NIH/q splenocytes ( P <0.05), in which the stimulation index (SI) of MLR was 3.51±0.55, and the footpad increment of DTH was (7.41±0.60)×10 -2 mm. NIH/q mice splenocytes injected via the systemic vein or third-party C57/BL mice splenocytes injected via the portal vein did not abolish the capability of the animals to develop MLR responses and DTH responses to NIH/q alloantigens ( P >0.05), in which the SI of MLR was 34.83± 3.90 and 35.33±2.54, the footpad increment of DTH was (23.51±1.69)×10 -2 mm and (23.87±2.76)×10 -2 mm ( P <0.05). Conclusions\ Portal venous inoculation of allogeneic cells could induce immunotolerance to alloantigens and this was donor antigen specific.\;
