摘要
目的:探讨羟基磷灰石(HA)缓释碱性成纤维细胞生长因子(bFGF)对停经后骨质疏松状态下植入体骨整合的影响。方法:20只大鼠双侧卵巢摘除术后3个月进行胫骨植入体植入术,再喂养3个月处死,样本进行显微CT和生物力学检测。结果:缓释bFGF导致BV/TV、%OI、Tb.N、Tb.Th等骨小梁微观结构参数值明显高于对照组,植入体最大推出力值和相对剪切力值亦高于对照组。结论:以HA为缓释载体局部应用bFGF能够增强停经后骨质疏松状态下植入体周围新骨形成、植入体-骨整合以及机械稳定性能。
Objective: To investigate the effect of control-released basic fibroblast growth factor(bFGF) from hydroxyapatite(HA) on bone-implant integration and implant fixation in ovariectomized(OVX) rats.Method: Three months after bilateral ovariectomy,20 rats were randomly assigned into two groups for tibiae implantation with or without control-released bFGF.At three months after implantation,all animal were sacrificed and the tibiae were harvested.Micro-CT was used to exam the trabecular microstructure and bone-implant integration,while push-out test was used to evaluate the mechanical fixation of these implants.Results: We found that the bFGF control-released from HA induced more peri-implant new bone formation in OVX rats,and the morphological parameters such as BV/TV,%OI,Tb.N and Tb.Th were higher than control levels,moreover,bFGF induced a marked increase in the maximal load and the interfacial shear strength.Conclusion: Controlled release of bFGF from HA may promote bone-implant integration and implant fixation following postmenopausal osteoporosis.
