不同硒水平对氟中毒大鼠的影响

摘要 目的:研究不同硒水平对氟中毒大鼠的影响。方法:雄性SD大鼠60只,随机分为6组,对照组饮蒸馏水饲普通饲料,实验组饮含氟(F-)45mg/L的蒸馏水,氟组饲普通饲料,4个氟硒组饲料中分别加入硒1.37mg/kg、1.6mg/kg、2.3mg/kg、4mg/kg,实验2个月。每周观察大鼠的一般情况,收集24h尿液,测定尿氟含量及对大鼠氟斑牙进行分级。分别于实验1月末和2月末每组8只大鼠采取尾血,测谷胱甘肽过氧化物酶和过氧化氢酶的活性。结果:实验组与对照组相比体重增长慢、尿氟含量升高、血清GSH-PX和CAT活性下降。4个氟硒组较氟组体重增长快、尿氟含量升高、氟斑牙症状减轻,GSH-PX、CAT活性升高。结论:适量硒能纠正自由基代谢紊乱,对高氟具有一定的拮抗作用,过量则对氟有协同作用,以2.3mg/kg硒对氟中毒的拮抗效果最好。 Objective: To study the effect of different selenium level on rats with fluorosis.Methods: Sixty male SD rats are divided randomly into 6 groups.The control group is fed with normal diet and distilled water.The experimental groups are fed with distilled water conatining 45m/L fluoride(F.).The fluoride group is fed with normal diet.Four fluorine and selenium groups are fed with diet containing selenium with concentration of 1.37mg/kg,1.6mg/kg,2.3mg/kg,4mg/kg.The experiment lasts two months,in which general conditions of rats is observed per week and 24-hour urine is collected to determine fluoride content in urine.The groups are then divided into several stages by the detal fluorosis.At the end of the first and the second month,8 rats in each group are selected to collect tail blood to measure serum glutathione peroxidase activity of catalase.Results: The growth rate in body weight of the experimental groups was lower than that of the control group.The fluoride content in urine of the experimental groups was highter than that of the control group.The activity of glutathione and catalase of the experimental groups was lower than that of the control group.The growth rate in body weight of four fluorine and selenium group was highter than that of the fluoride group.The fluoride content in urine of four fluorine and selenium group was highter than that of the fluoride group.The symptoms of dental fluorosis of four fluorine and selenium group were lighter than that of the fluoride group.The activity of glutathione and catalase of four fluorine and selenium group was highter than that of the fluoride group.Conclusion: The right amount Selenium can reduce free radical metabolic disorder,thus has certain antagonism effect on the fluoride,however,excessive selenium could have a synergistic effect of fluoride.The optimal concentration is 2.3mg/kg selenium.
