摘要
目的探讨黄连素对125I粒子放射治疗荷人肝癌裸鼠移植瘤的增敏效应。方法选取荷人肝癌HepG2细胞移植瘤的BALB/c雌性裸鼠24只,随机分成4组,每组6只。A组:生理盐水+空心125I粒子;B组:黄连素45mg/kg+空心125I粒子;C组:生理盐水+125I粒子;D组:黄连素45mg/kg+125I粒子。观察各组小鼠的肿瘤生长情况,计算黄连素增敏系数(EF),应用免疫组化法检测各组血管内皮生长因子(VEGF)和肿瘤组织微血管密度(MVD)的水平。结果免疫组化显示D组与其他组比较,VEGF和MVD的表达明显减弱,均P<0.05,且D组EF为1.84。结论黄连素可能通过抑制VEGF表达、改善肿瘤血管形成情况以增强肿瘤细胞的放射敏感性。
Objective To investigate berberine on the ^125I particle in radiosensitization and its effects of angiogenesis on hepatocellular carcinoma nude mice. Methods We successfully established model of liver cancer on 24 BALB/c female nude mice, and randondy divided into 4 groups (n =6): Group A, nude mice was given implanta- tion of blank particles and daily intragastric administration of NS; Group B, nude mice was given implantation of a blank particles and 45 mg · kg ^-1/d orally berberine solution; Group C, nude mice was given ^125I implantation of a particle (0.8 mCi) according to TPS; Group D, nude mice was given ^125I implantation of a particle (0. 8 mCi) according to TPS and 45 mg · kg-l/d orally berberine solution; The size of tumors were measured every 4 days and also the sensitivity coefficient were calculated. The expression of VEGF and MVD in different group was detected by immunohistochemistry. Results The expression of VEGF and MVD apparently decreased in the ^125I particles combined with berberine group(P 〈 0.05 ) compared with the other groups by immunohistochemistry, and the EF of berberine was 1.84. Conclusions Berberine has definite activity to enhancing the tumor radiosesitivity and the study indicate that berberine exert the effect by inhibiting the expression of VEGF and MVD.
