霍乱弧菌是引起霍乱的病源菌。霍乱弧菌产生的分泌性毒素CT是致病的主要物质之一。CT是多聚蛋白体,由A、B两个亚单位组成,A亚单位是毒性部分。
This paper is to report the subcloning of CT gene from a recombinant plasmid earring V.cholerae 79004 strain (Eltor biotype, isolated in China)CT DNA. The restriction mapping and nucleotide sequencing of the CT were performed. The cloned CT BamH Ⅰ fragment of the plasmid p551 was 21kb in size. A CT Pst Ⅰ fragment could be cloned from p551 into pBR322; the resulting recombinant plasmid was designated as pCT52. In an attempt to obtain the A^-B^+ in gene, Cla Ⅰ-Bgl Ⅱ fragment of pCT52 was inserted into the location between Cla Ⅰ-BamH Ⅰ sites of pBR322, such recombinant plasmids was pCB3. These clones, peT52, pCB3 were homologous with LT-B gene. A restriction mapping of pCT52 were obtained using common enzymes.Utilizing M13 cloning-chain termination sequecing method, the nucleotide of A2 and B subunits gene of 79004 was analysed. The sequencing deta showed that there is one nucleotide different from the data published by Lockman, H. et al., but no significant differece in gene structure. There might be no alteration in CT amino acid sequence by deducing.
