复方芪珠方对肝纤维化大鼠4种细胞因子表达的影响 被引量:5

Effect of Compound Qizhu Tablet on the Expression of 4 Cytokines in Rats with Liver Fibrosis

摘要 目的:观察复方芪珠方的不同提取物对大鼠肝脏肿瘤坏死因子(TNF-α)、白介素-4(IL-4)、白介素-8(IL-8)、白介素-10(IL-10)表达的影响,探讨复方芪珠方抗肝纤维化的作用机制。方法:70只大鼠随机分为空白、模型组、复方芪珠方水提高、低剂量组、复方芪珠方醇提高、低剂量组,秋水仙碱组。除正常组外,其余各组大鼠皮下注射40%四氯化碳(CCl4)橄榄油造模,首次5 mL.kg-1,以后每次3 mL.kg-1,每周2次,共8周。于造模开始次日给药,复方芪珠方高、低剂量组16.2,9.72 g.kg-1ig,秋水仙碱组0.5 mg.kg-1ig,每日1次,连续8周。造模结束后所有大鼠处死,肝脏组织学观察,测定肝脾指数,酶联免疫吸附试验(ELISA)技术检测TNF-α,IL-4,IL-8,IL-10在肝内的表达。结果:模型组大鼠肝组织炎症程度及纤维化程度均高于正常组,且肝组织中TNF-α,IL-4,IL-8的表达也明显增加,IL-10的表达则显著减少;应用复方芪珠方后,肝组织炎症和纤维化程度降低,同时相对模型组,肝组织中TNF-α,IL-4,IL-8的表达减少,IL-10的表达增加。结论:复方芪珠方用于大鼠肝纤维化有显著效果,其机制可能与其能影响TNF-α,IL-4,IL-8,IL-10等细胞因子在肝组织的表达有关。 Objective: To investigate the effect of Compound Qizhu tablet on the expression of tumor necrosis factor -a (TNF-a) , interleukin-4 (IL-4) , interleukin-8 (IL-8) and Interleukin-10 (IL-10) in rats with liver fibrosis. Method: Seventy SD rats were randomly divided into normal group, model group, Qizhu high-dose group (water preparation), Qizhu low dose group (water preparation), Qizhu high-dose group (ethanol preparation) , Qizhu low dose group (ethanol preparation) , and colchicine group. All the rats except normal group received 40% CC14 subcutaneous injection twice a week for 8 weeks, rats was firstly given 40% CC14 (5 mL· kg^(-1)) and 3 mL .kg-lsecondly. Rats in both Qizhu high-dose group and small dose group were ig given 16.2, 9.72 g ·kg^(-1) Qizhu decoction respectively, while rats in colchieine group were fed with 0.5 mg·kg^(-1) colchicine. Liver histology and liver and spleen index were observed after the modelling. The expression of TNF-a, IL-4, IL- 8 and IL-10 was detected by enzyme linked immune sorbent assay (ELISA). Result: The degree of hepatic fibrosis in all treatment groups was obviously reduced compared with model group. Compared with normal group, the expression of TNF-R, IL-4 and IL-8 in model group was elevated significantly, whereas the expression of IL-10 / was decreased (P 〈0.01 ). Compared with modelfgroup, the expression of TNF-a, IL-4 and IL-8 in treatment groups were decreased significantly, whereas the expression of IL-10 was increased (P 〈 0.01 ). Conclusion: Compound Qizhu tablet has obvious therapeutic effect on rats with liver fibrosis. The mechanism may be correlated to interfering the disturbance of some cytokines such as TNF-R, IL-4, IL-8 and IL-10.
