摘要
目的:研究FEN1基因在人食管癌中的mRNA和蛋白表达及其与肿瘤发生的关系。方法:采用SYBRGreen实时定量PCR技术检测32例食管癌组织及癌旁组织中的FEN1基因的mRNA水平;同时采用免疫组化技术检测上述32例组织中FEN1的蛋白水平及分布。结果:与癌旁组织相比,FEN1基因在食管癌组织中mRNA水平较癌旁组织显著上调,约为癌旁组织的3.46倍。免疫组化的结果也表明FEN1基因在食管癌组织中表达要高于癌旁组织。结论:FEN1基因在人食管癌中较癌旁组织表达显著上调。FEN1基因的表达与食管癌的发生密切相关。
Objective:To detect the expression of FEN1 in human esophageal squamous cancer and the relationship with tumor. Methods: Using SYBR Green real-time quantitative PCR and immunohistochemistry to detecte mRNA expression levels of FEN1 gene of 32 pairs of esophageal squamous cancer tissues and cancer adjacent tissues. Results: Comparing with cancer adjacent tissues, FEN1 gene mRNA levels in esophageal squamous cancer tissues significantly increased by 3.46 times. Immunohistochemistry also demonstrated that the mRNA expression levels of FEN1 gene in esophageal squamous cancer tissue was significantly up-regulated in relation to cancer adjacent tissues. Conclusion: Comparing with cancer adjacent tissues, the mRNA and protein level of FEN1 in human esophageal squamous cancer was significantly up-regulated. FEN1 gene expression may involved in esophageal squamous cancer development.
