摘要
目的探讨端粒酶基因(hTERC)扩增与宫颈上皮内肿瘤(CIN)和宫颈癌发生发展的关系及临床意义。方法应用荧光原位杂交技术检测100例存在宫颈病变患者的宫颈脱落细胞中hTERC基因的扩增情况。结果 100例各类宫颈细胞病变中,hTERC基因扩增比例分别为:CIN1组13.3%(4/30),CIN2组71.0%(22/31),CIN3组和宫颈癌组均为100%(19/19,20/20),各组比较差异均有统计学意义(P<0.01)。结论宫颈CIN和宫颈癌细胞hTERC基因均有扩增,且扩增率随病变的进展而增加;高级别CIN和癌的扩增率明显高于低级别CIN;检测hTERC基因扩增可以作为宫颈CIN和宫颈癌筛查及预后评估的重要指标。
Objective To study the relationship and clinical significance between human telomerase gene amplification and development of cervical intraepithelial neoplasm and cervical squamous cancer.Methods hTERC gene amplification were detected by FISH in cytological samples from 100 cases of CIN /cervical squamous cancer and 20 cases of normal cervix.Results In 100 cases of various types of cervical cell lesions,the abnormal amplification ratio of hTERC gene was:13.3%(4/30) in CIN1 group,71.0%(22/31) in CIN2 group,CIN 3 group and cervical cancer squamous group were 100%(19/19,20/20),each group compare difference to all have statistical significance(P 0.01).Conclusion hTERC gene was amplified both in CIN and cervical squamous cancer cells,and the amplification rate increases with lesion progression.The amplification rate of high grade CIN and squamous cancer was significantly higher than that of low grade CIN.Detection of hTERC gene amplification can be used as an important indicator in cervical CIN and cervical cancer screening and prognostic evaluation.
