摘要
目的探讨人表皮生长因子受体2(Her-2/neu)在胃癌组织中的表达及与胃癌生物学行为和预后的相关关系。方法采用SP免疫组化技术检测158例胃癌及其癌旁组织中Her-2/neu蛋白的表达,将检测结果与胃癌临床病理参数及患者随访资料进行相关性分析。结果 158例胃癌组织中,Her-2/neu蛋白阳性表达率为26.58%(42/158),癌旁正常胃组织中均未见Her-2/neu蛋白的阳性表达;Her-2/neu蛋白表达与肿瘤的浸润深度、病理分期、淋巴结转移、脉管神经受累及患者生存时间相关(P<0.05),但与患病年龄、性别、肿瘤大小及肿瘤部位无相关性(P>0.05)。Her-2/neu蛋白阳性表达的胃癌患者中位生存时间及生存率均明显低于Her-2/neu蛋白阴性者(P<0.05)。结论 Her-2/neu蛋白表达与胃癌生物学行为密切相关;检测Her-2/neu表达对胃癌临床预后评估及其治疗的选择有一定的指导意义。
Objective To investigate the association of the expression of human epidermal growth factor receptor 2 （Her-2/neu） and clinicopathologic variables and prognosis in patients with gastric cancer. Methods The expressions of Her-2/neu protein in 158 samples of gastric cancer（group A） and tumor-adjacent normal tissues（group B） were detected by immunohistochemical technique. The correlation of the Her-2/neu expression and elinicopathologic variables and overall survival was analyzed. Results The positive expression rate of Her-2/neu protein in group A was 26.58%, which of group B was zero. The positive expression of Her-2/neu protein was closely correlated to the depth of invasion, TNM stage, lymph node metastasis, vessel invasion and the overall survival（P〈0. 05）, but not correlated to the sex, age, tumor size and location（P〉0. 05）. The median survival time and survival rate were significantly lower in the patients with Her-2/neu positive than those with Her-2/neu-negative（P^0. 05）. Conclusion The expression of Her-2/neu protein is closely related to the biological behaviors of gastric cancer. The detection of Her-2/neu expression is helpful in evaluating the treatment and the prognosis of patients with gastric cancer.
