摘要
本文通过建立HBsAg的检测,对时间分辨荧光免疫分析技术进行了实验研究。采用单克隆抗体包被12孔微量滴定条,以二乙烯三胺五醋酸酐两步法将Eu^(3+)和单克隆抗体结合,制备示踪物。实验结果表明:本法简便,灵敏度较酶联法高,标记物有效期长,无污染环境等优点。
A time-resolved fluoroimmunoassay (trFIA) for human HBsAg is described, in which antibodycoated microtiter strip-wells and Eu^(3+) chelate-labeled monoclonal antibody are used. These Eu^(3+) ions bound to the antibody molecules by means of the anhydrid of diethylenetriaminepentaacetic acid. The solidphase antibody is immobilized microtiter strip-wells in which serum samples were assayed in two-step procedure. The precision, sensitivty and clincal studies indicate the assay is accurate. The trFIA is technically simple, and takes a count time of only one second. In addition, the Eu^(3+) label which is nontoxic presents no wast-disposal problems and has a long shelf life.
