摘要 目的建立人血浆中万古霉素的高效液相色谱测定法,为临床监测血药浓度以调整万古霉素的给药剂量提供方法学基础.方法以去甲万古霉素为内标,色谱柱为依利特Hypersil BDS C8(4.6 mm×250 mm,5μm),流动相为20 mmol/L的磷酸二氢钾-乙腈,采用梯度洗脱,流速为1.0 mL/min,检测波长为236 nm,柱温40℃.结果本测定法在1.313~105 mg/L间线性良好(r=0.999 1),日内、日间精密度≤5.2%,绝对回收率≥66%.结论高效液相色谱法测定万古霉素的血药浓度是一种快速可靠、结果准确的方法. Obejective To establish a high performance liquid chromatography(HPLC)method for the determination of concentration of vancomycin in human plasma.Methods Norvancomycin was used as the internal standard.The chromatography was performed on a Hypersil BDS C8(4.6 mm×250 mm,5 μm)column with the mobile phase consisted of 20 mmol/L potassium dihydrogen phosphate-acetonitrile,gradient elution was used,the flow rate was 1.0 mL/min,UV detection was set at 236 nm and column temperature was 40 ℃.Results The calibration curve was linear within the range of 1.313~105 mg/L(r=0.991).The intra-and inter-day precisions were less than 5.2%,the absolute extraction recoveries were more than 66%.Conclusion HPLC is a rapid,simple and accurate method for detecting the plasma concentration of vancomycin.
