摘要
以自交亲和芸芥7-5-1-1为研究材料,对影响芸芥花药愈伤组织诱导的主要因素进行了初步研究.结果表明:MS+2.0 mg/L 2,4-D+1.0 mg/L 6-BA+0.5 mg/L NAA是诱导芸芥花药愈伤组织适宜的培养基;芸芥花药在离体培养前最佳低温(4℃)预处理时间为2d,诱导率可高达46.2%;在诱导期间始终处于弱光或完全黑暗条件下的花药愈伤组织诱导率最高;单核期花药的愈伤组织诱导率最高,与单核前期、双核期花药的愈伤组织诱导率相比,差异达到极显著水平.
Using self-compatible Yunjie named 7-5-1-1 as the material, several main influencing factors on anther callus induction were studied. The results showed that the optimum medium for anther callus induction in Yunjie was MS+2.0 mg/L2, 4-D +1.0 mg/L 6-BA + 0.5 mg/L NAA. The optimum low-temperature pretreatment time was 2 days before Yunjie anther callus induction, and its callus induction rate could reach 46.2%. During inducting, anther callus induction rate under conditions of total darkness was the highest. There was obvious significance in callus induction between mono-nuclear anther and dual-nucleic anther and pre-nucleic anther.
