摘要
目的观察马钱子粗粉及其超微粉分别对雄性、雌性昆明小鼠的急性毒性反应,考察微粉化对其毒性的影响。方法一次性分别灌胃给予雄性、雌性昆明小鼠不同剂量的马钱子超微粉或粗粉,观察急性毒性反应,记录死亡潜伏期,用序贯法计算LD50及其95%可信限。结果马钱子超微粉雄性小鼠LD50为194.7 mg/kg,95%可信区间为208.7~180.7 mg/kg,马钱子超微粉雌性小鼠LD50为163.0 mg/kg,95%可信区间为198.2~127.8 mg/kg;马钱子粗粉雄性小鼠LD50为295.7 mg/kg,95%可信区间为331.7~259.7 mg/kg,马钱子粗粉雌性小鼠LD50为271.8 mg/kg,95%可信区间为323.7~219.9 mg/kg;粗粉的死亡潜伏期长于超微粉,而雄性动物的死亡潜伏期也长于雌性。结论马钱子超微粉及其粗粉对小鼠的毒性反应相似,其急性毒性存在量时效关系,但马钱子超微粉比粗粉的毒性大,临床应用马钱子超微粉应减少用量。
Objective To study acute toxicity and evaluate the safety of the crude powder and ultramicro powder of semen strychni on male and female Kunming mice, and to discuss the effects of micronization on the acute toxicity of semen strychni. Methods Male and female Kunming mice were given different doses of crude powder and ultramicro powder of semen strychni, the reaction symptoms of acute toxicity and death incubation periods were observed. The 50% lethal doses (LDso) and 95% confidence intervals of two kinds of powder of semen strychni were calculated by sequential trial. Results The LDs0 and 95% confidence intervals of semen strychni crude powder were 295.7 mg/kg and 323.7~219.9 mg/kg for male mice respectively, and 271.8 mg/kg and 323.7- 219.9 mg/kg for female mice respectively. The LD50 and the 95% confidence intervals of semen strychni uhromicro powder were 194.7 mg/kg and 208.7-180.7mg/kg for male mice respectively, and 163.0 mg/kg and 198.2-127.8 mg/kg for female mice respectively. The death incubation period of crude powder was longer than that of the ultramicro powder, and the death incubation period of male mouse was longer than that of female mouse. Conclusion The toxicity response of mice is similarity between ultramicro powder and crude powder of Strychni Semen. Their acute toxicity have a time-effect relationship in a dose dependent manner. The ultramicro powder of semen strychni is more toxic than the crude powder taking LD50 as index, so the ultramicro powder of semen strychni should reduce its dose in clinic.
