摘要
以柠檬酸钠为螯合剂,采用水热法合成了单分散的Eu3+掺杂NaGdF4纳米粒子,采用扫面电镜(SEM)、X射线粉末衍射(XRD)、荧光光谱对粒子进行了表征,结果表明,所合成的纳米粒子尺寸在60-80nm之间,粒度均匀,且具有良好的发光性能。基于其荧光性质,将其标记于肺癌细胞表面,利用激光扫描共聚焦显微镜对标记后的细胞进行荧光成像。在488 nm激发下,获得了红色荧光图像,48h后细胞仍然保持着很亮的荧光,显示出荧光纳米粒子很好的荧光稳定性,并采用MTT法检测肺癌细胞的存活率,对纳米粒子的毒性进行了初步评估。
The monodispersed NaGdF4:Eu nanoparticles were synthesized via a hydrothermal approach. Citric acid rare-earth was used as the precursor. The properties of as-prepared nanoparticles were characterized by scanning electron microscopy( SEM), X-ray diffraction( XRD), particle size analyzer, fluorescence spectra. Experimental results showed that the nanoparticles were single crystal hexagonal phases with an average size of around 60 nm,exhibiting excellent fluorescent characteristics. Based on their fluorescence properties, NCI-H460 Lung cancer cells were labeled by the prepared nanoparticles, and were studied by using laser confocal micro- scope. Red fluorescence images were collected under the excitation of 488nm. After 48h the cells still remained red fluorescence, that showed the nanoparticles had good photochemical stability. And the cell toxicity of the nanoparticles were detected through The MTT method
