摘要
为研究Kisspeptin-10(Kp-10)对禽类生殖内分泌的影响,将75羽22日龄雌鹌鹑随机分为3组,分别腹腔注射300μL生理盐水(对照组),注射0.1 nmol Kp-10(低剂量组)和1 nmol Kp-10(高剂量组),每日注射1次,连续注射3周。记录鹌鹑开产情况,并于60日龄时采集血液和卵泡组织,测定生殖激素与受体的mRNA表达。结果:与对照组相比,注射Kp-10组鹌鹑产蛋率显著提高,且大卵泡数显著增加,血液中雌激素水平显著升高;低剂量组输卵管重显著增加,但体重、肝脏重和小卵泡重无显著变化。Real-time PCR结果显示,Kp-10处理显著下调小卵泡及F1级卵泡上促黄体激素受体(LHR)、促卵泡激素受体(FSHR)和促性腺激素释放激素Ⅰ(GnRH-Ⅰ)的基因表达。
In order to investigate the effect of Kisspeptin-10 (Kp-10) on birds, 75 female quails (22 days) were subjected to 0 (control), 0. 1 nmol (low dosage) or 1.0 nmol (high dosage) Kp-10 administration via intraperitoneal injection daily. The administration was lasted for 21 days. Kp-10 was dissolved in saline and the total volume for injection was 300 p.L. The information of egg production was recorded daily. At 60 days, quails were sacrificed and samples including blood and ovarian follicle were collected for reproductive hormones and receptor mRNA analysis. The results showed that, compared with control group, egg productions were significantly increased in low or high dosage of Kp-10 treatment groups, and there were more numbers of large ovarian follicles. Quails in low dosage group also showed a significantly higher weight of oviduct. However, Kp-10 did not change the body weight or the weight of small follicles or livers. Real-time PCR results showed that the luteinizing hormone receptor ( LHR), follicle stimulating hormone receptor (FSHR) and gonadotropin releasing hormone receptor (GnRHR) mRNA expression levels in F1 follicles and small follicles were significantly down-regulated after Kp-10 administration.
