摘要
为了探讨β-酪啡肽-5(β-CM5)对大鼠小肠葡萄糖吸收的影响,利用翻转的离体小肠模型,通过测定60 min内β-CM5不同浓度组肠囊内葡萄糖含量及葡萄糖的转运率;测定离体肠道黏膜α-葡萄糖苷酶、Na+-K+-ATP酶活力,分析大鼠小肠钠依赖性葡萄糖共转运载体(SGLT-1)及葡萄糖转运载体2(GLUT-2)mRNA表达水平,揭示β-CM5对葡萄糖吸收影响及机制。结果:不同浓度β-CM5肠囊内葡萄糖的转运量及转运率均显著低于对照组;β-CM5组小肠黏膜中α-葡萄糖苷酶和Na+-K+-ATP酶活力显著低于对照组;β-CM5组小肠黏膜中SGLT-1、GLUT-2 mRNA表达量与对照组相比均显著性下调。结果表明:β-CM5可以通过抑制葡萄糖吸收的相关酶活性,下调葡萄糖转运载体mRNA的表达,从而抑制肠道葡萄糖的吸收,提示β-CM5可能具有降糖作用。
This study is to explore the influence of β-CM5 on the absorption of glucose in intestine using the reverted sacs of adult rats in vitro. The glucose concentration and transport velocity of glucose in the different concentrations of β-CM5 groups within 60 minutes were detected. In addition, the activities of α-glucosidase and Na+-K+-ATPase were examined and the mRNA expression levels of sodium/glucose transporter 1 (SGLT-1) and glucose transporter 2 (GLUT-2) were analyzed. The results showed that glucose contents in intestinal capsule volume, transfer rate, Na+-K+-ATPase and α-glucosidase activities in β-CM5 groups were significantly lower than those in the control group. Compared with the control group, SGLT-1 and GLUT-2 mRNA expression levels of β-CM5 group were significantly decreased. The results indicated that β-CM5 can inhibit the activities of enzymes related to glucose uptake and reduce the mRNA expression of glucose transporter, and thus inhibit intestinal absorption of glucose. It suggested that β-CM5 might have a hypoglycemic effect.
