摘要
目的建立鸡胚致死孤儿病毒PCR检测方法,并应用于鸡胚及禽源性生物制品的检测。方法根据鸡胚致死孤儿病毒长纤突保守序列分别设计3对引物,优化并建立鸡胚致死孤儿病毒PCR检测方法;分别采用该方法对SPF鸡胚和流感疫苗、麻疹疫苗进行检测。结果建立的方法可检测到10-9稀释的病毒DNA;该方法与鼠腺病毒及猴腺病毒无交叉反应;在SPF鸡胚及禽源性生物制品中应用该方法均未检出病毒。结论该方法具有良好的特异性和敏感性,可以应用于禽源性生物制品中鸡胚致死孤儿病毒的检测。
Objective To establish the PCR method for chicken embryo lethal orphan virus(CELOV),and apply it in avian-derived biological product.Method Three pairs of primer are designed according to the long fiber(LF) gene of CELOV;establish the PCR method using these primers.The SPF chicken eggs,influenza vaccine and measles vaccine were detected by the PCR method respectively.Result The CELOV DNA can be detected by this method when it was diluted to 10-9;there is not cross reaction in CELOV,MAdV and SAdV detection.And this DNA was not detected in SPF chicken eggs and avian-derived biological products.Conclusion This PCR method has nice specificity and sensibility;it can be applied in detecting of CELOV in SPF chicken eggs and avian-derived biological products.
