染色体消除法诱导啤酒大麦单倍体技术研究 被引量:1

Study on Inducting Barley(Hordeum vulgare) Haploids Using Hordeum Bulbosum Chromosome Elimination in H.Vulgare × H.Bulbosum Crosses

摘要 为了探索完善染色体消除法诱导啤酒大麦单倍体的技术体系,以7个啤酒大麦F1代材料为母本,分别以1个二倍体和1个四倍体的球茎大麦为父本进行杂交诱导。结果表明,二倍体球茎大麦的单倍体诱导率平均为28.13%,四倍体球茎大麦的单倍体诱导率为0。研究了试验期间的环境日最高温度、不同授粉时间和授粉后激素处理对授粉结实率的影响,结果表明,杂交试验期间环境日最高温度低于22℃、啤酒大麦去雄后2d进行授粉、授粉后第1~3d每天用Dicamb∶2,4-D∶GA3=1∶2∶75(质量比,各激素浓度分别为1、2、75mg·L-1)的混合激素喷授粉小花1次,可获得最高的杂交结实率。同时,分析了不同年际间授粉结实率存在较大差异的原因和不同倍性的球茎大麦对单倍体诱导率的影响。 Seven varieties of H.vulgare as maternal parents were crossed with a diploid H.bulbosum and a tetraploid H.bulbosum to induce haploids in their progeny , diploid H.bulbosum as male parents has on average 28.13% of the pollinated florets formed embryos , and the tetraploid H.bulbosum is 0. In the article, the effects of pollination time, daily maximum temperature during the pollination, and spraying exogenous hormones after pollination on seed setting rate is analysed. The result shows the highest seed setting rate will be gained by pollinating when two days after manual emasculation, spraying the mixed hormones of Dicamb∶2, 4 D∶GA3=1∶2∶75 once a day during the first three days after pollination, and controlling the maximum temperature in the day during the pollination period less than 22 ℃. The reason of difference of seed setting rate between the year 2010 and 2011, effects of H.bulbosum genetype on haploid inducing rate are discussed.The objective of this article is to provide basic knowledge and practice guidelines in barely (Hordeum vulgare ) haploids breeding.
