摘要
目的 :采用贝美格腹腔注射制作大鼠急性癫持续状态 ( SE)模型 ,观察海马神经元中 bcl- 2、bax、p5 3的表达。 方法 :通过将 SD大鼠脑海马切片行 bcl- 2、bax、p5 3的免疫组化染色 ,观察肿瘤相关基因在大鼠 SE模型海马中的作用。 结果 :bcl- 2在正常大鼠海马中不表达 ,发作高峰期在 CA 3区有弱表达 ;bax在正常大鼠海马中弱表达 ,在发作高峰期 CA3区强表达 ;p5 3(突变型 )在正常大鼠海马及 SE模型中均不表达。 结论 :急性癫持续状态可造成海马神经元的凋亡 ,其部位主要位于 CA 3、CA1区 ,而且肿瘤相关基因 bcl- 2、bax在 SE过程中发挥作用 ,而p5 3(突变型 )
Objectives: Make the status epilepticus(SE)model in rat by celiac injection of Bemegride, observe the expression of bcl2, bax and p53 in hippocampus. Methods: Immuno histological method was used to show the expressions of tumorrelated genes bcl2, bax and p53 in hippocampus of SD rats. Results: bcl2 did not express in normal rat hippocampus, slightly expressed in CA3 region during SE; bax slightly expressed in normal rat hippocampus, strongly expressed in CA3 region during SE; mutated p53 did not express in normal and SE rats. Conclusions: SE can cause apoptosis of hippocampal neurons, which mainly located in CA3 and CA1 regions. Tumorrelated genes bcl2 and bax take part in the process of SE, while mutated p53 does not.
