摘要
目的:探讨胰腺癌组织中X染色体连锁凋亡抑制因子相关因子1(XAF1)基因启动子区甲基化状态及蛋白的表达。方法:采用甲基化特异性聚合酶链反应(MSP)及免疫组化SP法检测胰腺癌组织(48例)、胰腺良性疾病组织(26例)和正常胰腺组织(15例)中XAF1基因启动子区甲基化状态及蛋白的表达。结果:胰腺癌、胰腺良性疾病及正常胰腺组织中,XAF1基因启动子区甲基化率分别为31.3%、7.7%和0%,XAF1蛋白的阳性表达率分别为52.1%、84.6%及88.9%,3组比较差异均有统计学意义(P=0.005;χ2=11.169,P=0.004)。XAF1基因启动子区甲基化状态及蛋白的表达均与胰腺癌的TNM分期、分化程度及淋巴结转移关系密切(P<0.05)。胰腺癌组织中XAF1基因启动子区甲基化状态与蛋白表达呈负关联(rP=-0.463,P<0.001)。结论:XAF1基因启动子区甲基化是该基因失活的重要机制之一,XAF1甲基化修饰水平在胰腺癌的发生、发展中起重要作用。
Aim :To study the relationship between status of methylation of X-linked inhibitor of apoptosis protein asso-iated factor-1 ( XAF1 ) gene promoter and expression of its protein in pancreatic carcinoma. Methods : Methylation-specific PCR and immunohistochemical SP technique were used to detect the status of methylation of XAF1 gene promoter and the protein expression in pancreatic carcinoma( n = 48 ), benign pancreatic disease ( n = 26 ) and normal tissue ( n = 15 ). Re- sults: In normal tissue,there was no promoter methylation of XAF1 gene, while in pancreatic carcinoma and benign pancre- atic disease,the methylation rate were 31.3% and 7.7%. The positive expression rate of XAF1 protein in pancreatic carci- noma,benign pancreatic disease and normal tissue was 52.1% ,84.6% and 88.9% ,and there were significant differences (P =0. 005 ;X2 = 11. 169, P = 0. 004). The methylation status and expression of XAF1 protein were related to the tumor TNM stage, pathological grade and lymph node metastasis (P 〈 O. 05). There was correlation between methylation of XAF1 gene promoter and protein expression in pancreatic carcinoma tissue ( rp = - 0. 463,P 〈 0. 001 ). Conclusion : Methylation of promoter might be one of the important factors of XAF1 gene inactivation, and it might play an important role in carcino- genesis and progression of pancreatic carcinoma.
