不同照射剂量对肝癌细胞生长抑制率的影响 被引量:1

摘要 目的探讨不同照射剂量对肝癌细胞生长抑制率的影响。方法对SMMC 7721肝癌细胞,应用国产X射线深部治疗机进行照射,肝癌细胞共分为7组,分别给予0Gy(假照组)、0.5Gy、1Gy、2Gy、4Gy、6Gy、8Gy进行照射。通过进行细胞克隆形成实验计算细胞存活分数。采用AnnexinⅤ和PI双染,然后采用流式细胞术检测不同辐射剂量对肝癌细胞早期凋亡的影响。结果细胞克隆形成实验结果表明,在不同剂量0、0.5、1、2、4、6和8Gy的照射组中,细胞存活分数分别为89.12±1.43、84.17±1.58、80.45±2.35、73.21±2.56、70.23±3.05、67.48±2.72、61.38±1.79%。与假照组(0Gy照射组)比较,其他照射组的细胞存活分数均明显低于假照组(P<0.05)。与0.5Gy和1Gy组相比,2、4、6、8Gy照射组的细胞存活分数明显减低(P<0.05)。而0.5Gy和1Gy照射组之间无显著差异。2、4、6Gy组之间比较无显著差异。而8Gy照射组的细胞存活分数明显低于任何其他组(P<0.05)。在不同剂量0、0.5、1、4、6和8Gy照射组中,细胞凋亡率分别为5.13±0.24、6.34±0.43、8.16±0.41、10.47±0.53、14.38±0.26、18.57±0.44和22.39±0.36%。与对照组相比,各实验照射组中细胞凋亡率均显著高于对照组(P<0.05),各组间比较均存在显著差异(P<0.05)。结论随着照射剂量的增加细胞凋亡比率明显增加,细胞存活分数减少。因此照射剂量是影响细胞生存率的重要因素。 Objective To explore the influence of the different dose of irradiation on human hepatoma cell growth inhibition rate.Methods Smmc7721 were irradiated with domesitic deep X ray therapeutic instrument and the irradiation group can be divided into 7 groups:0Gy(sham-radiation group)、0.5Gy、1Gy、2Gy、4Gy、6Gy、8Gy group.Cell fraction can be calculated with cell colony forming experiment.Use annexin V and PI double staining and then use flow cytometry to detect the influence of different dose of irradiation on human hepotoma cell early apoptosis.Results The conclusion of cell colony forming experiment showed that at different doses of irradiation group,that is 0Gy(sham-radiation group)、0.5Gy、1Gy、2Gy、4Gy、6Gy、8Gy,cell survival fraction were 89.12±1.43、84.17±1.58、80.45±2.35、73.21±2.56、70.23±3.05、67.48±2.72、61.38±1.79%.Compared with sham-radiation group,cell survival fraction of the other group was significantly lower than sham-radiation group(P﹤0.05).Compared with 0.5Gy group and 1Gy group,2、4、6、8Gy radiation group have a significant reduction(P﹤0.05).While there was no statistically significant difference between 0.5Gy group and 1.0Gy group,as well as 2、4、6Gy group.Cell survival fraction of 8Gy group obviously lower than the other group.At different radiation group,that is 0(sham-radiation group)、0.5、1、2、4、6、8Gy group,apoptosis rate were 5.13±0.24、6.34±0.43、8.16±0.41、10.47±0.53、14.38±0.26、18.57±0.44和22.39±0.36%.Appoptosis rate of each experimental group was obviously higher than control group(P〈0.05).Comparision of each group exist significant differences(P﹤0.05).Conclusion With the increase of irradiation dose,apoptosis ratio increased significantly and the cell survival fraction decrease.Thus irradiation dose is an important factor in impact of cell survival rate.
