摘要
为了提高稻米中生育酚的含量,本研究经农杆菌介导将由CaMV35S启动子驱动的水稻尿黑酸植基转移酶基因(OsHPT)导入粳稻品种武育粳3号,经RT-PCR等分析证明导入基因能在水稻胚乳中表达。HPLC测定结果表明,水稻中过表达OsHPT后,转基因稻米胚乳中总生育酚的含量极显著高于未转化对照,且主要表现为α-生育酚含量的提高,总生育酚和总维生素E含量最高可分别提高1.5和0.8倍。
In order to increase the tocopherol content of rice, the homogentisate phytyltransferase (OsHPT) gene was transferred into an elite rice japonica cultivar Wuyujing3 driven with CaMV 35S promoter by Agrobacterium-mediated transformation RT-PCR analyses confirmed the expression of the input transgene in rice endosperm. HPLC results showed that after OsHPT was expressed in the rice, the total tocopherol content in endosperm of transgenic rice lines was significantly higher than the wild type, mainly for the increase in α-tocopherol Content, In transgenic rice, the contents of total tocopherol and vitamin E increased maximally 2.5 and 1.8 folds respectively compared with those of wild type.
