药物干预治疗对动脉粥样硬化合并牙周炎兔基质金属蛋白酶的影响

Effect of drug intervention on the expression of matrix metalloproteinase-2 in rabbits with atherosclerosis and periodontitis

摘要 目的观察阿洛伐他汀联合替硝唑对动脉粥样硬化合并慢性牙周炎兔大血管及心肌组织中基质金属蛋白酶-2(matrix metalloproteinase-2,MMP-2)表达的影响。方法 48只动脉粥样硬化合并慢性牙周炎的雄性新西兰大白兔随机分成无药物干预组(A组)与药物干预组(B组)各24只,第16~20周进行药物灌胃,A组给予生理盐水,B组给予阿托伐他汀片+替硝唑片,20周末处死动物取样本,采用免疫组织化学及组织病理HE染色方法检测2组血管和心肌组织中MMP-2水平。结果 B组血管及心肌组织中MMP-2水平均低于A组(P<0.05);A组血管MMP-2表达量低于心肌组织(P<0.05);B组血管MMP-2表达量高于心肌组织(P<0.05)。结论阿洛伐他汀联合替硝唑可降低动脉粥样硬化合并慢性牙周炎兔动物模型MMP-2水平,可能是防治动脉粥样硬化合并慢性牙周炎的方法之一。 Objective To observe the influence of atorvastatin and tinidazole on the expression of matrix metalloproteinase-2 (MMP-2) in myocardial tissue and large blood vessels of rabbits with atherosclerosis and chronic periodontitis. Methods Forty-eight male New Zealand white rabbits with atherosclerosis and chronic periodontitis were randomly divided into group A (without drug intervention) and group 13 (intervention group). Group A was given normal saline and group B was given atorvastatin+tinidazole by gavage 16 to 20 weeks later. All the animals were sacrificed and taken samples after 20 weeks. The expression of MMP 2 in blood vessels and myocardial tissue was determined with immunohistochemistry and HE staining. Results The level of MMP-2 in blood vessels and myocardial tissue in group B was lower than that in group A (P〈0.05), and was higher in blood vessles than that in myocardial tissue in group B (P〈0.05). Conclusion Atorvastatin+tinidazole can reduce the level of MMP-2 in the rabbit model with atherosclerosis and chronic periodontitis, which may be one of the methods for preventing and treating atherosclerosis and chronic periodontitis.
