摘要
以加拿大红枫(Acer rubrum L.)的顶芽和侧芽为外植体,考察不同消毒剂处理时间对消毒效果的影响,以及植物生长调节剂及其浓度配比对愈伤组织诱导及不定芽增殖的影响。结果表明,外植体的最佳采集时期为4月中旬;先用体积分数70%的乙醇处理15 s,再用1 mg/mL HgCl2处理7 min,最后用20mg/mL NaClO浸泡25 min,加拿大红枫外植体的消毒效果好,且存活率最高,为90%。诱导加拿大红枫愈伤组织的适宜培养基为1/2 MS+2.0 mg/L 6-BA+0.5 mg/L 2,4-D+0.1 mg/L NAA+0.1 mg/L GA3。较适合加拿大红枫不定芽增殖的培养基为1/2 MS+1.0 mg/L 6-BA+0.1 mg/L NAA+0.1 mg/L GA3。
Using the terminal buds and lateral buds of Canadian maple(Acer rubrum L.) as explants,the effect of disinfectant treatment time on disinfection effect and plant growth regulators on callus induction and adventitious bud proliferation was studied.The results showed that the best collecting period of explants was in mid-April.The optimum disinfection effect could be obtained by treating in 70% alcohol for 15 s,then in 1 mg/mL HgCl2 for 7 min and in 20 mg/mL NaClO for 25 min.The optimal medium for inducing callus was 1/2 MS+2.0 mg/L 6-BA+0.5 mg/L 2,4-D+0.1 mg/L NAA+0.1 mg/L GA3.The optimal medium for adventitious bud proliferation was 1/2 MS+1.0 mg/L 6-BA+0.1 mg/L NAA+0.1mg/L GA3.
