复方银黄微型灌肠剂对腹腔巨噬细胞5-脂氧合酶产物生成的影响

EFFECT OF YINHUANG MICRO-ENEMA COMPOUND ONTHE SYNTHESIS OF 5-LIPOXYGENASE PRODUCTS IN RATS

摘要 目的:研究复方银黄微型灌肠剂(Yinhuang Micro-enema Compound,YHMCE)对5-脂氧合酶(5-LOX)产物生成的影响,以阐明其抗炎作用的分子机制。方法:体外培养大鼠腹腔巨噬细胞(Peritoneal macrophage,PM),采用高效液相色谱法测定白三烯B4(LTB4)和5-羟二十碳四烯酸(5-HETE)含量变化,以反映5-LOX活性变化。结果:在生药浓度2.24~8.96g/L的范围内,YHMCE呈剂量依赖方式抑制大鼠PM LTB4、5-HETE的生物合成,其IC50分别为含生药2.41g/L与8.43g/L。结论:YHMCE通过抑制5-LOX活性,减少LTB4及5-HETE的产生,可能是其发挥抗炎作用的分子作用机制之一。 Objective: To study the effect of Yinhuang Micro-enema Compound (YHMCE) on the synthesis of 5-1ipoxygenase products and elucidate its anti-inflammatory mechanisms. Methods: Peritoneal macrophage (PM) obtained from normal rats was cultivated in vitro. The levels of leukotrienes B4 (LTB4) and 5- hydroxy-eicosatetraenoic acid (5-HETE) were detected with reversed phase high-performance liquid chromatography (RP-HPLC) to reflect the activity of 5-1ipoxygenase (5-LOX). Results: YHMCE between 2.24 g/L and 8.96 g/L could inhibit the synthesis of LTB4 and 5-HETE with dose-dependent manner in rat peritoneal macrophage. The 50 % inhibiting concentration (IC50) was 2.41 g/L and 8.43 g/L ; respectively. Conclusion: The inhibitive effects of YHMCE on the activity of 5-LOX, LTB4 and 5-HETE may account for its anti-inflammatory actions.
