摘要
【目的】优化提取荷叶多糖的最佳工艺参数及研究其抗氧化作用,为荷叶多糖的提取和利用提供技术参考。【方法】以荷叶为原料,采用单因素试验及正交试验设计,从提取时间、提取温度、料液比等方面,对荷叶多糖的提取工艺进行优化;并研究荷叶多糖对羟基自由基(·OH)和超氧阴离子自由基()的清除作用。【结果】热水浸提荷叶多糖的最佳工艺条件为:浸提时间2.0h,浸提温度75℃,固液比1∶30。荷叶多糖对·OH和均有明显的清除作用,且清除率随多糖浓度增加而增大,其最高清除率分别为51.6%和23.1%。【结论】在最佳提取工艺条件下,荷叶多糖的提取率为9.95%;荷叶多糖抗氧化作用明显。
[ Objective ]The aim of this experiment was to optimize parameters of extracting polysaccharide in lotus leaf and study its antioxidation in order to provide technical references for extraction and utilization of polysaccharide in lotus leaf. [ Method ]Single factor experiment and orthogonal experiment were designed to optimize extracting technology for polysaccharide in lotus leaf from extraction time, extraction temperature, and material/liquid ratio to study the scavenging effects of polysaccharide on hydroxyl radical and superoxide radical. [Result]Optimal extraction parameters of polysaccharide in lotus leaf using hot water extraction were as follows: extraction temperature of 75℃, extraction time of 2.0 hours, and material/liquid ratio of 1:30. Polysaccharides in lotus leaf had strong scavenging effects on hydroxyl radical and superoxide radical with the highest scavenging rate of 51.6% and 23.1%, respectively. Further, scavenging rate increased as polysaccharide concentration increased. [Conclusion]Under the best extraction conditions, extraction rate of polysaccharide in lotus leaf was 9.95%, and polysaccharide in lotus leaf had strong antioxidation.
