风轮菜活性部位对大鼠离体胸主动脉的舒张作用及机制研究 被引量:2

Vasorelaxant Effect and Mechanism of Active Fraction from Clinopodium chinense on Isolated Thoracic Aorta of Rats

摘要 目的研究风轮菜活性部位(CCE)对大鼠离体胸主动脉的舒张作用及其可能的作用机制。方法采用离体大鼠胸主动脉张力实验,观察CCE的舒张血管作用,并观察CCE对大鼠胸主动脉产生NO的影响。结果 CCE显著降低苯肾上腺素(PE,10-6mol/L)与高钾(60 mmol/L)预收缩内皮完整血管的张力,对PE诱导血管收缩的抑制作用强于对高钾的作用。但是,CCE对去内皮的主动脉环均无舒张作用;一氧化氮合酶(NOS)抑制剂N-硝基-L-精氨酸甲酯(L-NAME)或吲哚美辛存在的情况下,CCE抑制PE收缩的效应消失。另外,CCE可明显增加大鼠主动脉NO含量。结论CCE对大鼠胸主动脉产生内皮依赖性舒张作用,该作用与促进NO的合成释放有关,也可能与促进前列环素的生成及抑制受体操控性钙通道和(或)肌浆网Ca2+释放有关。 Objective To investigate the vasorelaxant effect of active fraction from Clinopodium chinense(Benth.) O.Ktze(CCE) on isolated thoracic aorta of rats and its mechanism.Methods The isotonic contraction of the thoracic aorta strips in rats was recorded,and the effects of CCE on the contraction of rat thoracic aorta were examined.In addition,thoracic aorta rings were incubated with different concentration of CCE,nitric oxide(NO) level was measured.Results CCE evoked a significant relaxation in endothelium-intact aorta rings precontracted by phenylephrine(PE,10-6mol / L) and KCl(60 mmol / L).But,the effect on endothelium-intact aortas induced by PE was more effective than that induced by KCl.Additionally,CCE had no significant effect on endothelium-denuded aorta rings precontracted by phenylephrine and KCl.Pretreatment with nitric oxide synthase(NOS) inhibitor Nw-nitro-L-arginine methyl ester(L-NAME),or indomethacin,vasodilatation effect of CCE was remarkably reduced.Furthermore,thoracic aorta rings pretreated with CCE,NO level was significantly increased.Conclusion These results indicate that CCE possesses the endothelium-dependent relaxation which is probably related with promoting synthesis and release of NO and prostacyclin(PGI2),as well as the inhibition of receptor-operate Ca^2 + channel and(or) release of Ca^2 + from sarcoplasmic reticulum.
