孔圣枕中丹对SHR大鼠左右脑DAT、DRD_1和DRD_2基因的影响 被引量:5

The Effect of Kongsheng Zhenzhong Pill to the DAT、DRD1 and DRD2 Gene Expression on the Left and Right Brain of SHR Rat

摘要 目的探讨孔圣枕中丹对ADHD动物模型SHR大鼠左右侧大脑半球前额叶皮质、纹状体的DAT、DRD1和DRD2基因表达的影响。方法把30只SHR大鼠随机分为三组,分别用孔圣枕中丹、盐酸哌甲酯和生理盐水灌胃28 d,采用荧光定量PCR检测其左右脑前额叶皮质、纹状体的DAT、DRD1和DRD2基因的表达。结果在灌服孔圣枕中丹复方煎液后,SHR大鼠前额叶皮质和纹状体右侧的DAT和DRD1基因表达比左侧高,纹状体右侧的DRD2基因表达比左侧高,差异有统计学意义(P<0.05),前额叶皮质右侧的DRD2基因表达比左侧高,但差异无统计学意义。结论孔圣枕中丹可能主要通过调控右侧前额叶皮质-基底神经节环路的DA神经信号传导而治疗ADHD。 Objective To investigate the effect of Kongsheng Zhenzhong Pill to the DAT、DRD1 and DRD2 gene expression on the left and right prefrontal cortex、corpus striatum brain of ADHD animal model.Methods 30 SHR rats were randomly divided into three groups.Each group was gavaged with Kong-sheng Zhenzhong Pill,methylphenidate hydrochloride and saline for 28 days,rspectively.Using fluorescence quantitative PCR detected the left and right prefrontal cortex、corpus striatum DAT、DRD1 and DRD2 gene expression.Results Kongsheng Zhenzhong Pill group’s DAT and DRD1 gene expression on the right of prefrontal cortex and corpus striatum was higher than the left,DRD2 gene expression on the right of corpus striatum was higher than the left with significant statistical difference(P 〈 0.05).DRD2 gene expression on the right of prefrontal cortex was higher than the left without significant statistical difference(P 〉 0.05).Conclusion Kongsheng Zhenzhong Pill can cure ADHD by regulating the right of the prefrontal cortex-basal ganglia loop of the DA nerve signal transduction to achieve.
