肉碱对大鼠力竭运动后心肌线粒体呼吸链功能及抗氧化能力的影响 被引量:5

Effects of exogenous carnitine on function of respiratory chain and antioxidant capacity in mitochondria of myocardium after exhaustive running in rats

摘要 目的:探讨肉碱对大鼠心肌线粒体呼吸链功能及抗氧化能力的影响。方法:40只2月龄雄性Wistar大鼠随机均分为4组(n=10):对照组(C)、补充肉碱组(LC)、运动训练组(T)、运动训练+补充肉碱组(TLC)。LC和TLC组每天按300 mg/(kg bw.d)剂量灌胃补充肉碱一次,T组和TLC组进行递增负荷水平跑台运动训练,共6周。力竭运动后即刻取样,差速离心法提取线粒体。分光光度法测定线粒体呼吸链CⅠ-Ⅳ、超氧化物歧化酶(SOD)活性及丙二醛(MDA)含量。结果:与C组相比,LC组、T组和TLC组CⅠ及CⅣ活性均显著升高(P〈0.05,P〈0.01),T组和TLC组CⅡ及CⅢ活性均显著升高(P〈0.05,P〈0.01);与LC组相比,TLC组CⅠ-Ⅳ活性均显著升高(P〈0.05,P〈0.01);与T组相比,TLC组CⅠ及CⅣ活性均显著升高(P〈0.05)。与C组相比,LC组、T组和TLC组SOD活性均显著升高、MDA含量均显著降低(P〈0.05,P〈0.01);与LC组和T组相比,TLC组SOD活性均显著升高、MDA含量均显著降低(P〈0.05)。结论:补充肉碱及运动训练均可提高心肌线粒体呼吸链功能及抗氧化能力,从而提高机体的运动能力,且肉碱与运动训练具有协同作用。 Objective: To investigate the effect of exogenous carnitine on function of respiratory chain and antioxidant capacity in mitochondria of myocardium in rats.Methods: Forty male Wistar rats were randomized to 4 groups(n=10): static control(C),supplementation of carnitine(LC),exercise-training(T) and training with supplementation of carnition(TLC).LC and TLC animals were perfused carnitine by the dose of 300 mg/kg bw·d.T and TLC animals were forced to performed 6-week treadmill training.Heart were prepared immediately after exhaustive running.Myocardium mitochondria was extracted by differential centrifugation.Spectrophotometric analysis was used to evaluate activities of respiratory chain complex(C) Ⅰ-Ⅳ and superoxide dismutase(SOD),malondialdehyde(MDA) level in myocardium mitochondria.Results: To compare with C group,CⅠand CⅣ activity in LC,T and TLC group were increased significantly(P〈0.05,P〈0.01),CⅡ and CⅢ activity in T and TLC group were increased significantly(P〈0.05,P〈0.01);to compare with LC group,CⅠ~Ⅳ activity in TLC group were increased significantly(P〈0.05,P〈0.01);to compare with T group,CⅠand CⅣ activity in TLC group increased significantly(P〈0.05).To compare with C group,SOD activity increased remarkably,MDA was remarkably lower(P〈0.05,P〈0.01) in LC,T and TLC group;To compare with LC and T group,SOD activity increased remarkably,MDA was remarkably lower(P〈0.05) in TLC group.Conclusion: Carnitine and training could improve function of respiratory chain and increased antioxidant capacity in myocardium mitochondria,there was better function of cooperation between carnitine and training.
