人乳腺癌MCF-7细胞系中肿瘤干细胞的富集与鉴定 被引量:1

摘要 采用无血清培养液(SFM)悬浮细胞聚球法体外培养人乳腺癌MCF-7细胞系,第5、10、15天拍照,观察干细胞球的生成。悬浮培养第5天可以开始观察到悬浮细胞球形成,细胞球形成效率为(0.6±0.5)%,第10天时细胞球形成效率为(2.4±1.2)%,比第5天时明显增多,培养第15天时细胞球形成效率达到(3.8±1.8)%,MCF-7悬浮成球细胞中侧群细胞(SP)细胞含量为(3.9±1.4)%,高于常规培养的MCF-7贴壁细胞(1.1±0.5)%(P<0.05),采用无血清悬浮细胞聚球培养法可以从MCF-7细胞系中简便、高效地富集乳腺癌干细胞。 MCF-7 cells were suspension cultured in vitro with a serum-free medium (SFM), and sphere-forming ceils were observed and taken pictures were at the 5 th, 10 th, and 15 th day. The typical cell spheres could be observed after 5 days with the proportion of sphere-forming cell (0. 6 + 0. 5 ) %. The proportion of spheres-forming cell was(2.4 + 1.2)% in 10 days, which was significantly higher than that in 5 days and reached(3.8 + 1.8)% in 15 days. Side population (SP) cells in the MCF-7 floating sphere-forming ceils were significantly higher than those of the MCF-7 adherent ceils, which was (3.9 + 1.4)% vs(1.1 + 0. 5)% (P 〈0. 05). By suspension culture method with SFM, the breast cancer stem ceils in MCF-7 cell line can be isolated and enriched conveniently and effectively.
