β3肾上腺素能受体激动剂BRL37344对3T3-L1前脂肪细胞分化的影响 被引量:4

Effect of β3-adrenoceptor agonist BRL37344 on differentiation of preadipocyte 3T3-L1 strain

摘要 目的观察β3肾上腺素能受体激动剂BRL37344对3T3-L1前脂肪细胞分化及相关基因mRNA转录的影响,并探讨其抗肥胖的作用机制。方法将3T3-L1前脂肪细胞分为4组:空白对照组、常规分化组、BRL37344处理组、常规分化+BRL37344组。采用油红O染色法初步判断各组脂肪细胞的分化情况,RT-PCR法检测各组脂肪细胞分化过程中过氧化物酶体增殖物激活受体γ2(Peroxisome proliferators activated receptorγ2,PPARγ2)、脂肪细胞型脂肪酸结合蛋白(Adipocyte fatty acidbinding protein,aP2)、脂蛋白脂肪酶(Lipoprotein lipase,LPL)基因mRNA的转录水平。结果经不同浓度的BRL37344处理8 d后,3T3-L1前脂肪细胞中PPARγ2、aP2、LPL基因mRNA的转录水平均下调,且呈剂量依赖性,其中10-7mol/L BRL37344作用明显;在不同的分化时间点,经10-7mol/L BRL37344处理后,细胞分化相关基因与相应对照组相比,均呈下调趋势(P<0.05)。结论β3肾上腺素能受体激动剂BRL37344可能通过下调脂肪细胞分化相关基因的转录,抑制3T3-L1前脂肪细胞的分化。 Objective To observe the effect of β3-adrenoceptor agonist BRL37344 on the differentiation of preadipocyte 3T3-L1 strain and investigate the action mechanism of BRL37344 against corpulence.Methods 3T3-L1 preadipocytes were divided into four groups,i.e.blank control,basic inducer,BRL37344 and BRL37344 + basic inducer groups.The differentiation of adipocytes in various groups were judged by oil red O staining,while the transcription levels of peroxisome proliferators activated receptor(PPARγ2),adipocyte fatty acid binding protein(aP2) and lipoprotein lipase(LPL) mRNAs by RT-PCR.Results The transcription levels of PPARγ2,aP2 and LPL mRNAs in preadipocyte 3T3-L1 strain 8 d after treatment with BRL37344 decreased significantly in a dose-dependent mode.The down-regulatory effect of BRL37344 at a concentration of 10-7 mol / L was satisfactory.However,all the mRNA levels of differentiation-related genes in preadipocyte 3T3-L1 strain at various time points after treatment with 10^-7 mol / L BRL37344 showed decreasing tendencies as compared with those in the corresponding control groups(P〈0.05).Conclusion β3-adrenoceptor agonist BRL37344 might inhibit the differentiation of preadipocyte 3T3-L1 strain by down-regulating the transcriptions of adipocyte adipocyte differentiation-related genes.
