摘要
【目的】探讨麝香配伍冰片对脑外伤模型大鼠脑组织含水量、水通道蛋白-4(AQP-4)表达的影响机制。【方法】雄性大鼠随机分为假手术组、模型组和麝香配伍冰片组,用Feeney法造成大鼠脑挫裂伤模型,干湿重法测定脑组织含水量,并采用免疫组化方法检测脑组织AQP-4蛋白的表达。【结果】模型组大鼠伤后2 d伤侧脑组织含水量、伤灶周围AQP-4蛋白表达水平均明显高于假手术组(P<0.05);麝香配伍冰片治疗组脑组织含水量、AQP-4表达水平均较模型组降低(P<0.05)。【结论】麝香配伍冰片可以明显减轻创伤后脑水肿,其作用可能与抑制AQP-4在损伤脑组织中的表达有关。
[Objective] To explore the effects of moschus combined with borneol on cerebral edema and expression of AQP-4 in injuried rats and discuss the possible mechanisms. [Methods] The rats were randomly divided into sham operation group, traumatic brain injury (TBI) group and moschus combined with borneol group. Parietal brain contusion was made by Feeney' s weight-dropping method. Brain water contents was measured by wet-dry weight and immunohistochemistry technique was used to determine the protein expression of AQP-4. [ Results]The brain water contents and AQP-4 protein expression of TBI group were obviously higher than those of the sham operation group (P〈 0.05). In mosehus combined with borneol treatment group, the brain tissue water content and the expression of AQP-4 were obviously lower than those of the model group (P 〈 0.05). [ Conclusion ] Moschus combined with borneol can obviously alleviate edema volume, which may be related with the inhibiting the expression of AQP-4 protein.
