摘要
X -射线辐照枳橙 ( 2n =2x =18)叶肉原生质体 ,碘乙酸处理Page橘柚 ( 2n =2x =18)悬浮细胞系原生质体 ,电场诱导两种原生质体融合 ,经培养再生出愈伤组织和胚状体。对再生的愈伤组织进行细胞学检查 ,观察到二倍体 ( 2n =2x =18,52 .2 % )、四倍体( 2n =4x =36,5.8% )和非整倍体细胞 ( 4 2 .0 % )。选取OPA0 7、OPE0 5、OPAN0 7和OPW0 1等 4种随机引物分析胚状体的杂种特性。RAPD检测结果说明所得到的胚状体均为通过非对称融合得到的枳橙和Page橘柚的体细胞杂种。
X-irradiated mesophyll protoplasts of citrange(Poncirus trifoliata×Citrus sinensis)were electrofused with embryogenic protoplasts of Page tangelo(C.reticulata×C.paradisi),which were treated with 0.25mmol/L iodoacetic acid for 15 min.Calli and embryoids recovered from the fusion products.A lot of diploid and aneuploid cells,together with few tetraploid cells,were observed.Four arbitrary primers,OPA07,OPE05,OPAN07 and OPW01,were selected to identify the hybridity of the embryoids.The results of RAPD analyses showed that the em- bryoids were somatic hybrids between Citrange and Page tangelo.
