摘要
目的 观察银杏黄酮对大鼠离体心脏缺血再灌注损伤的保护作用。方法 采用离体大鼠心脏Langendorff灌流 ,低灌缺血 40min ,再恢复常速灌流 30min ,造成心肌缺血再灌流损伤。利用该模型观察银杏黄酮对缺血再灌流心肌心缩力 (ST)、左室内压 (LVP)、心率 (HR)、冠脉流出量 (CBF)、冠脉流出液中乳酸脱氢酶 (LDH)、心肌丙二醛 (MAD)含量及超氧化物歧化酶 (SOD)活性的变化。结果 银杏黄酮 15mg/L和 30mg/L可改善缺血再灌注所致心肌收缩力及左室内压的降低 ,减少LDH释放量及心肌MAD生成量 ,并提高SOD活力。结论 银杏黄酮对离体心脏缺血再灌注有保护作用 ,其作用与直接清除自由基外 ,亦可能与SOD活性的维持有关。
Objective The protective effect of ginkgo biloba flavone on isolated heart isochemia reperfusion injury in rats was observed. Methods Isolated langendorffs heart of rats were used. The Model of acute heart isochemia reperfusion injury was Produced by hypoperfusion for 40min followed up reperfusion for 30min. Systolic tension(ST), left ventricle pressure(LVP), heart rate(HR), coronary blood flow(CBF), lactate dehydrogenase(DHE) from CBF, and malondialdehyde(MDA) and superoxide dismutase(SOD) of cardiac muscle were determined. Results Ginkgo biloba flavone 15mg/L and 30mg/L improved the ST and LVP that reduced by reperfusion, decreased the release of LDH and the content of MDA in cardiac muscle, and increased the activity of SOD. Conclusions Ginkgo biloba flavone possesses protective effect on isolated heart ischemia reperfusion injury. The machanism magbe indudes the remaining of SOD activity in cardiac muscle except the free radicals were scavenged directly.
