摘要
目的 :探讨温阳散结中药的抗肿瘤机制 ,研究温阳散结中药诱导BEL 740 2细胞凋亡作用。方法 :采用细胞形态学、原位末端标记和流式细胞技术 ,观察温阳散结中药对BEL 740 2细胞的影响。结果 :BEL 740 2细胞经中药处理后 ,光镜下可见凋亡细胞的形态学特征性改变 ,TUNEL标记阳性 ,流式细胞仪分析DNA含量 ,在非凋亡细胞二倍体峰 (G1)之前细胞出现典型的凋亡细胞峰 (Ap峰 )。 0 0 0 5g·ml-1中药组和 0 0 1g·ml-1中药组发生凋亡的细胞数分别为 13 9%和 2 1 6 %。结论 :温阳散结中药可诱导BEL 740 2细胞发生凋亡 ,并有随浓度升高作用增强的趋势 ,实验结果表明 ,诱导肿瘤细胞凋亡可能是温阳散结中药治疗肝癌的作用机理之一。
Objective:To investigate the antitumor mechanism of Chinese berbs for warming yang and dispersing stasis and observe the action of these herbs in inducing apoptosis of BEL 7402 cells. Method:Apoptosis of BEL 7402 cells induced by the aboved said herbs was detected through fast red tablet staining,flow cytometry and terminal uridine deoxynucleotidyl and labeling (TUNEL). Result:After treating BEL 7402 cells with the herbs typical apoptosis characteristics could be seen under light microscope and TUNEL positive cells were detectable.The apoptotic rate was 13.9%(drug concentration 0.5%) and 21.6% (drug concentration 1%)Conclusion:Chinese herbs for warming Yang and dispersing stasis could induce apotosis of BEL 7402 cells and this effect depends on drug concentration. Inducing the apoptosis of tumor cells may be part of the mechanism displayed by Chinese herbs for warming Yand and dispersing stasis in treating hepatoma. [
