摘要
目的 探讨基质金属蛋白酶抑制剂 - 2 (TIMP - 2 )对SGC - 790 1胃癌细胞体外侵袭能力的影响。方法 将人全长TIMP - 2基因经脂质包裹转染PA317细胞系 ,G418筛选 ,感染人SCG - 790 1胃癌细胞系 ,经体外细胞增殖、细胞电泳率、软琼脂形成和侵袭小室的实验综合分析其生物学行为改变。结果 经TIMP - 2基因转染的SGC - 790 1细胞和电泳率、软琼脂形成和侵袭能力均明显低于对照组 ,而与对照组相比细胞增殖曲线没有明显的变化。结论 TIMP - 2基因转染可以使肿瘤细胞的表面电荷发生变化 ,使细胞的电泳能力、细胞的移动性和侵袭穿透能力减弱 ,而与细胞和生长周期没有明显影响。提示TIMP -
Aim:To invastigate the in vitro change of invason of gastric carcinoma cell SGC-7901 transfected with matrix metalloproteinase-2(TIMP-2)gene.Methods:A full human TIMP-2 cDNA gene was constructed into virus vehicle PT-MT,and was transferred into human gastric carcinoma cells SGC-7901 and positive clone was established by Boyden chamber in vitro.Results:The expression of TIMP-2 gene in SGC-7901-T2 cells was higher than that in the SCG-7901 cells in vitro and the ability of invasion in Boyden chamber was weak in vitro.The cell proliferation curves of TIMP-2 transfected and TIMP-2 untransfected groups had no obvious difference.Conclusion:The TIMP-2 gene may be an important negative factor in gastric carcinoma cell invasion and metastasis.
