摘要
目的探讨不同饮食状态对实验性糖尿病肾病大鼠尿蛋白检测结果的影响。方法将清洁级健康雄性SD大鼠72只随机分为实验组(n=45)和对照组(NC组,n=27),实验组大鼠一次性腹腔注射链脲佐菌素(STZ)。两组大鼠分别在第4、8、12、16周留取正常饮食和禁食状态下24h尿液采用放免法检测尿蛋白,同时检测肾脏功能和形态。结果糖尿病大鼠成模率为73.1%,并出现肾小球肥大和肾小球基底膜增厚等肾脏病理学改变。实验组大鼠进食和禁食状态下不同时间点尿蛋白、体质量和血糖与NC组比较,差异均有统计学意义(P<0.05)。实验组大鼠禁食后不同时间点饮水量和尿量与NC组比较,差异均无统计学意义(P>0.05)。不同时间点尿蛋白与肾脏功能检测指标无关。结论禁食状态下糖尿病肾病大鼠尿蛋白与对照组比较,差异无统计学意义(P>0.05)。尿蛋白不能动态反映糖尿病肾病早期功能和形态学变化。
Objective To explore the influence of different dietary condition on urinary albumin(UAlb) level in rat model with diabetic nephropathy(DN).Methods 72 specific pathogen free(SPF) male Sprague-Dawley(SD) rats were randomly divided into control group(NC group,n=27) and experiment group(n= 45),which were induced with intraperitoneal injection of streptozotocin for once.At 4,8,12 and 16 weeks after injection,24 h urine of mice,under normal diet and absolute diet,in both groups were collected and measured for UAlb levels by immunoturbidimetry,and functional and morphologic changes of kidney were examined.Results The rate of developed diabetes mellitus(DM) model in SD rats,with pathologic changes of renal glomerulus hypertrophy and glomerular basement membrane thickening,was 73.1%.There was significant difference of urinary protein level,body weight and blood glucose level between mice in experiment group under normal diet or absolute diet,detected at different time,and mice in NC group(P〈0.05),but no statistical difference of volume of drinking and urine,detected at different time,could be demonstrated between the two groups(P〉0.05).There was no correlation between levels of urinary protein and kidney function at different time(P〉0.05).Conclusion Absolute diet could not affect urinary protein level in rat with DN.Urine level of protein could not dynamically inflect the functional and morphologic changes of kidney at early stage of DN.
