Smad2蛋白在子宫颈癌中的表达及意义

摘要 目的探究Smad2蛋白在正常子宫颈(normal cervical epithelium,NCE)、子宫颈上皮内瘤变(cervi-cal intraepithelial neoplasia,CIN)与子宫颈浸润癌(invasive carcinoma of cervix,ICC)中的表达情况,以及其与各种临床病理指标间的相关性。方法采用免疫组织化学SP法检测15例NCE、62例CIN和41例ICC组织石蜡样本中Smad2蛋白的表达情况,并结合子宫颈癌的临床病理指标进行分析。结果 Smad2蛋白主要定位在细胞胞质中,也有少量位于胞核中。Smad2蛋白在ICC中表达率为36.6%(15/41),与CIN9(42/62,67.7%)和NCE(13/15,86.7%)相比明显偏低(P<0.05)。对41例子宫颈癌样本的分析发现,Smad2蛋白阳性表达与子宫颈癌患者的年龄、组织学类型、临床分期、有无淋巴结转移无关(P>0.05),但与病理分级有关(P<0.05)。结论 Smad2蛋白在子宫颈癌组织中表达较正常组织和良性病变组织低,并与患者病理分级密切相关,Smad2蛋白表达的异常可能在子宫颈癌的发生与恶化中起到重要作用。 [Objectives] To investigate Smad2 expression in normal cervical epithelium (NCE), cervical intraepithelial neoplasia (CIN), invasive carcinoma of cervix (ICC) and to explore the potential correlations between Smad2 expression and clinicopathologic features of cervical carcinoma. [Methods] Smad2 protein level in 15, 62, 41 cases of NCE, CIN and ICC respectively were investigated with immunohistochemistry, [Resuits] Smad2 protein level was observed in cytoplasm dominantly, and in nucleolus slightly. Positive Smad2 expression was revealed in 36.6% (15/41) of ICC, lower than those of CIN (42/62, 67.7%) and NCE (13/15, 86.7%), significantly (P〈O.01). Based on the analysis of 41 ICC specimen and their clinicopathological data, a significant association between Smad2 protein level and histological grades (P〈0.05) was found, but no significant association between Smad2 protein level and other clinicopathological factors including ages, pathological types, clinical stages, lymph node metastasis was observed. [Conclusion] Smad2 has a lower protein level in cervical carcinoma tissue than in normal cervical epithelium and benign lesion epithelium, and it also has a strong association with histological grades in cervical carcinoma. Smad2 may play an important role in carcinogenesis and progression of cervical carcinoma.
