摘要
目的 探讨胶质瘤细胞血小板源生长因子 (PDGF)BB自分泌环活性与肿瘤细胞egr 1基因表达的关系 ,以及它们在胶质瘤发生、发展中的作用。方法 用mRNA原位杂交和免疫组化染色观察了 73例人脑胶质瘤组织。结果 胶质瘤细胞PDGFBmRNA表达水平随肿瘤恶性程度增加而升高。egr 1mRNA、EGR 1蛋白、PDGFα受体和β受体及酪氨酸磷酸化蛋白阳性肿瘤细胞密度彼此间均呈显著性正相关 ,并均随肿瘤恶性程度及肿瘤细胞PDGFBmRNA表达水平升高而同步递增。结论 胶质瘤细胞普遍存在PDGFBB自分泌环活性和egr 1基因表达异常增加 ,它们可能互为因果 ,并产生一个放大效应 ,这也许是胶质瘤发生、发展的重要因素。
Objective To investigate the relationship between activity of autocrine loop of platelet derived growth factor(PDGF) BB and expression of egr 1 gene in human gliomas, and their effect on development and progression of the gliomas.Methods 73 human gliomas with different grades of malignancy were studied by in situ hybridization and immunohistochemistry methods.Results The expression levels of PDGFB mRNA in glioma cells increased with the malignancy. The positive cell densities of egr 1mRNA, EGR 1 protein, α receptor and β receptor of PDGF BB, and phosphotyrosine protein correlated positively with one another in gliomas. All of them were elevated parallely with increasing of the malignancy and PDGFB mRNA expression level in the tumor cells. Conclusions There are generally abnormal enhancement of activity of PDGF BB autocrine loop and overexpression of egr 1 gene in glioma cells. They are possibly cause and result each other as well as produce a magnified effect. This may be the important factor of development and progression of human gliomas.
