p16基因对脑胶质瘤细胞作用的实验研究 被引量:1

摘要 目的 探索 p16基因在脑胶质瘤发生发展过程中的作用及 p16基因在脑胶质瘤临床诊断、治疗中的应用前景。方法 采用脂质体转染的方法 ,将外源野生型 p16基因导入胶质瘤细胞株 U2 5 1、C6 ,筛选阳性克隆。同时以空载体质粒 p CDNA3为对照 ,免疫组化检测 p16基因表达 ,用MTT法测定瘤细胞生长曲线及对转染的瘤细胞在裸鼠体内形成瘤块的变化进行分析。结果 转染 p16基因的 U2 5 1、C6细胞有外源 p16基因的整合及表达 ,克隆形成率减少 ,生长速度明显减慢 ,瘤细胞在裸鼠中形成瘤块体积显著降低。结论 导入外源野生型 p16基因可抑制胶质瘤细胞恶性增殖。 Objective To determine the effects of p16 gene on human glioma cell lines. Methods p16 gene was transfected into U251 and C6 glioma cell lines by lipofectin. Integration and expression of exogenous p16 gene were detected. MTT colorimetric assay was used to measure cell growth. Tumor mass formation of glioma cells was observed in nude mice. Results Expression of p16 gene was demonstrated by immunohistochemistry. The growth rate of both p16 gene transfected U251 and C6 reduced Tumor mass formation of tumor cells in nude mice was inhibited by transfected p16 gene. Conclusion The expression of p16 gene can inhibit the growth of glioma and tumor formation in nude mice. $$$$
