菊明降压方中总黄酮成分的提取工艺优选 被引量:1

Optimization of Extraction Technology for Total Flavonoids from Juming Jiangya Prescription

摘要 目的:优选菊明降压方中总黄酮成分的提取工艺条件。方法:以浸膏得率、总黄酮和蒙花苷含量为考察指标,紫外分光光度法测定总黄酮含量,HPLC测定蒙花苷含量,选取提取溶剂、溶剂用量、浸泡时间、提取时间和提取次数等为考察因素,采用单因素试验和正交试验法优选提取工艺。结果:最佳提取工艺条件为用80%乙醇浸泡15 min后提取3次,分别加8,6,6倍量80%乙醇各提取1.5 h。结论:优选的提取工艺条件合理、稳定,适用于实际大生产。 Objective: To optimize extraction technology of total flavonoids from Juming Jiangya prescription. Method: With yield of extract, the content of total flavonoids and buddleoside as indexes, thecontent of total flavonoids and buddleoside were determined by UV spectrophotometry and HPLC, respectively. extraction solvent, solvent dosage, soaking time, extraction time and extraction times were selected as investigating factors, extraction technology was optimized by single factor test and orthogonal test. Result: Optimized extraction conditions were: soaked 15 min with 80% ethanol, extracted 3 times with 8, 6, 6 times the amount of 80% ethanol, 1.5 hours for each time. Conclusion: Optimized extraction process was rational, stable and could be used in large-scale production.
